I'm basically the same person I was before I started my phd, only difference is the things I have gone through and learnt throughout this phd; for example I learnt a music software (of which I would never have done if I had not left my country and come here to do my phd!); and I started eating chicken....

first of all, from the response of the principal investigator, he has asked you to apply online like everybody else.
So you have the option to do that.

2nd, if he contacts your supervisor, most likely your supervisor might ask you, "And how many hours a week would you put into this new job--if you got it? Would you still be able to cope with your current phd work?"

3rd, should you ask your supervisor about the job, and see if anyone has contacted him---if you have already applied online, you could tell your supervisor (if you wanted) that you made a job application. It will look better for you that at least your supervisor is not the last person to find out about this (especially if you secured the job!).

4th, if you know someone who knows him, I dont know which "him" you are referring to. I assume it is the principal investigator. Well, if you know someone who knows "him", then a good idea would be if you could secure a letter of recommendation from this "person who knows him" and include that in your ONLINE application.

5th, is the job already taken? I don't know and since there is still a month before the closing date, I wouldn't worry about it if I were you. The post is just to fill forms---the truth is people want to do things quickly and efficiently; if they ask you to fill in forms, there must be a very valid reason for them to want you to fill up forms. They wouldn't want applicants to fill up forms for nothing.

to quantify the "brightness" of your band (how intense it is) instead of visual comparison with ladders then you need something like Genetools software. http://www.genegnome.com/html/products.html
Or gelQuant http://www.ampl.com.au/gq_frames.htm.

I haven't used open source applications for this. There are lots of other free apps but these are mostly for gene sequencing. http://molbiol-tools.ca/molecular_biology_freeware.htm

What you need to do is to go to the molecular labs in your university, ask the students there what software they are using to quantify their bands, and ask if you can have access to it. If you need to get permission from their supervisors, then ask first.

You only have 6 gels, this will be very fast. I don't think anyone will turn you down. Usually people in labs help each other. Ask someone to help you, make friends and ask for a suitable time so that this person may show you how to do it for one gel, and then you can do the other 5 gels. Its not like you have 600 gels.

Usually the software will be in the computer near the camera system used to photograph the gels. Genetools can be run on a mac but as I said, if your university only uses PCs, it is better to run the analysis on a lab in the computer and then just use the data on your mac.

Molecular labs usually will have this kind of software. Go and ask tomorrow. good luck! :-)
