Horray for research!

Hi all

You know when your project is pootling along well and interesting things happening - do you get all excited?

Its not very often but when I have times like that I get really happy and realise I love doing what I do and remember why I started on this journey.

Am I just a wierdo / saddo? We all tend to share on here when things aren't very good but I just wanted to say today that I think we are all quite lucky.

I know that feeling, I love that feeling. When I get colonies on my transformation plates, bands on my PCR gel, lots of ACGT's in my sequencing...all wonderful, beautiful things that make your heart glad you started your PhD. If I could sing, I would. Hugs and kisses to the world!!!!

I'm doing my first PCR this afternoon, I hope I get bands then I will be happy too!

It didn't work and I don't know if it was me doing something wrong or just the primers not working. I'm going to try again today with different primers. *fingers crossed*

katq, I am no PCR expert (far from it). But if I have a PCR that doesn't work the first time, I always try temperature, Mg etc gradients before I try different primers. If you have taken this into account already, ignore my advice and try other primers. Good luck, and have a beer for me when you get product!

I got it to work, well there were bands just not the right bands - but better than nothing. And I'm also finding all these incubation times are brilliant for coffee breaks! I'm starting to like Molecular Biology.

Don't you just hate it when stuff goes wrong when it's not your fault. I concentrated really hard on all my PCR and sequencing, and then I find out that the company sent the wrong clone in the first place so it isn't right anyway! Not a good start!
