First of all, I appreciate your progress in six months.
If your supervisor is very supportive, you can tell him that you are struggling to explain the results or whatever part of the article it is.
Whenever I wrote articles, discussions with my adviser helped me to complete the articles.

Help - rewriting intorduction and getting blocked

First of all, I appreciate your efforts to pursue ph.d. I have had 6 years of break in total between my degrees. It has been very difficult path that I came across.
I am also writing my thesis right now. Writing introduction is not that easy! It took me more than 1.5 months to formulate the introduction chapter of my thesis which is only 5 pages. But I directly started to write chapter 3 of my thesis. My supervisor is supportive. If I do not know how to formulate a particular section of a chapter, I ask openly and get an useful suggestion from my supervisor. May be you could ask your supervisor about his/her expectation and how you need to structure the introduction.
You are very strong, do not give up because of this temporary problem.

Major Revisions - 6 weeks and not heard back

When I submitted an article to a very good journal of my research area, they took 4 months to say that it was a major revision. Then it took another 4 months to get a response about acceptance of the article in its current form. I would say that not to worry much.

First first authored publication - aiming too low or over ambitious!?

If your supervisor is sort of expert in the particular field, then he/she can not go wrong. Besides that, this publication is an added advantage.

From my experience, one has to carefully choose the journal to submit the article. I have an experience of an article being rejected after 5 revisions, then we had to submit to different journal, but another article was accepted after one major revision.

Mine is also multi-disciplinary topic. It involves many hours of spending in lab. So, I try to work (such as reading, writing) during weekends, sometimes late nights or early mornings. I have been spending in lab during day time.

Have you asked your supervisor to suggest another methodology? If your supervisor really support you, there is nothing wrong in following his suggestion and make modification.
